Pete Bone's current jobs
Perform studies, using in-house core technology, to show levels of stabilisation at time intervals and at temperatures to be agreed upon depending upon the specific target and/or project. Technical Skills and Responsibilities: • Perform virus quantification assays (TCID50) • Freeze drying techniques using a Virtis Advantage Freeze Dryer • Maintenance of cell lines • Successful research into freeze dried excipients improving the ‘cake’ quality and aesthetics for client needs. Additional: In charge of company purchasing provided an opportunity to improve my computing skills by designing software allowing a more simplified purchasing process for everyone.
Pete Bone's past jobs
Providing molecular support to the Hepatitis C group through Mode-Of-Action assays, medium throughput compound screening and development of chimeric replicons from mutagenic studies. Technical Skills and Responsibilities: DNA/RNA: • Site/multi-site directed DNA mutagenesis • In-Fusion PCR cloning • Transformations, Minipreps and Midipreps to and from competent cells respectively • Performing PCR and QPCR • DNA sequencing (Beckman Sequencer) • Bioinformatics analysis of DNA sequences • RNA extraction and analysis • Producing in vitro RNA transcripts Cell Culture: • Maintenance of numerous cell lines • Routine screening of either replicons or compounds using luciferase and/or ELISA assays to test their activity • Routine mycoplasma testing • Transient transfection by electroporation of mammalian cells and interpreting results obtained via Activity Base analysis software Additional: Role provided me with the opportunity to work closely with the medicinal chemistry group, gaining insight into small molecule drug designing and organic chemistry methods.
Organising and managing the fermentation department. The fermentation procedure has involved me in a number of technical processes, ranging from working with live micro-organisms in the fermenters and cell lysis to the purification of proteins using FPLC, SDS PAGE, PCR and QPCR. In addition, I have used the molecular techniques of mini-preps and cell transformations.
The main objectives with this role involved working with the Bio-Enzyme Laboratory Team, Production Laboratory Team and Stock controller for efficient production and packaging of bio-enzyme reagents to ensure customer requirements are met, in accordance with GLP. Additional duties I have undertaken include the complete overseeing of Dye Terminator Removal Kit orders involving Sephadex preparation, aliquoting, packaging, spot checks and PCR quality control of each media batch number made. I have had the opportunity to improve on a number of skills within abgene including PCR, QPCR, gel electrophoresis and buffer preparation.
