1. Designed, synthesized small hybrid molecule library as neuroprotective compounds for optic neuropathy, ischemic brain stroke and Alzheimer's Disease targeting antioxidant pathway. 2. Designed fluoroscent probe with cell penetrating attribute to help finding mechanism of action for glucocorticoid receptor pathway. 3. Collaborated with biologists and clinicians, formulated, prepared grant proposals to NIH, DOD and funding institutions. 4. Designed series of small molecule natural product libraries and prodrugs as iNOS/NFK-b inhibitor for disease targeting inflammation, immunomodulation and angiogenesis. 5. Wrote a book chapter on PPAR pathway invited by Wiley. Manuscript accepted. 6. Taught graduate and undergraduate chemistry lectures and labs.

COMPANY OVERVIEW: AyuVis Research LLC is a near-clinical stage drug development company focused on improving the lives of patients in Intensive Care Unit (ICU) by, providing therapy to increase patient survival. The primary focus of AyuVis Research is to help patients with hyper inflammation and immune compromised diseases. The first indication for which AyuVis is seeking FDA approval is Bronchopulmonary dysplasia, a chronic lung injury of pre-term infants. DEVELOPMENT: We have identified and screened a library of small molecule, non-toxic, natural products, and established the possible mechanism of action. Lead compounds AVR-25, AVR-48 and AVR-123 were identified with demonstrated potent activity in vitro and in vivo mouse model. AyuV-25 and AVR-48 protected mice from lethal gram negative sepsis against Escherichia coli. Pseudomonas aeruginosa and CLP model of sepsis and will be developed for treating lung infection and acute respiratory distress syndrome. AVR-48 is being developed to prevent BPD in preterm babies and we received both Orphan Drug and Rare pediatric disease designation from the FDA. Our goal is to refine and extend our initial Structure Activity Relationship work and optimize for IV drug delivery in case of sepsis. We will complete in vivo testing of the analogs in higher animal models to provide a clinical candidate; and file an Investigational New Drug (IND) application for clinical trial. We have two approved patents on this proprietary technology in US and several other countries. AyuVis has been awarded SBIR grants from NIH Expertise and services: Provides consulting on pre-clinical drug discovery (lead optimization, Medicinal chemistry SAR), drug development, SBIR/STTR grant proposal writing, patent drafting, writing, compiling pre-IND, NDA, and 510(k) regulatory documents. Managed two medicinal chemistry projects and moved the projects from hit-to-lead and lead optimization stage in ophthalmology, Monitored and evaluated the project status to proceed to a go/no go decision. . Designed, synthesized back-up compound in antiviral program which progressed to preclinical study. Developed a scalable asymmetric synthesis of the lead anti-viral compound. New target proposition, structure based drug design, supervision of associate scientists. Assisted the CMC group in authoring, reviewing, editing CMC technical documents, pre-IND and NDA for FDA filing in 3 projects.

• Designed, synthesized SAHA and analogs as HDAC inhibitors to support glaucoma and retinal angiogenesis program. Identified and synthesized literature Tyrosine Receptor Kinase inhibitors with potent in vitro activity, improved aq. solubility desired for intra-vitreal dosing with significant in vivo anti-angiogenic activity for wet AMD program. • Synthesized the appropriate pro-drug of the VEGF-2 inhibitor and nitric oxide donor with improved aq. solubility and stability in retina and glaucoma program. • Identified, synthesized and optimized highly receptor selective 5-HT2A agonist with benzazepine core structure for serotonergic program in glaucoma with IOP lowering activity in monkey eyes. • Designed and synthesized novel and potent compounds as 5-HT2A receptor agonists in benzodifuran series serotonergic program in glaucoma to address the undesired systemic and CNS side effects. • Identified the lead bradykinin agonist compound which progressed to phase-I proof of concept clinical study for lowering intraocular pressure in glaucoma.
