Directed a team of separation technicians, and provided them guidance on method development and purification of complex synthetic molecules, biomolecules and other natural products. Simultaneously, independently purified the complex mixture of reaction products (from work-up) as well as natural products, by employing advanced techniques such as Prep HPLC (Reverse phase, C18 columns) and analytical HPLC method development as well as using Biotage Automated Flash Purification Systems. Selected appropriate HPLC column and solvent system for best separation. Chiral columns for separation of enantiomers. Independently and single handedly purified impure/ crude synthetic products (From chemical reactions or isolated from natural sources), using automated purification machines such as Biotage Selekt Flash Purification System, Teledyne NextGen CombiFlash machines and Prep HPLC. Used both normal phase and reverse phase (C18 columns) columns/ methods of purification. Solvent system modification using organic acids/ bases, to improve or develop the purification method for Prep HPLC and Biotage purifications. Spotting and handling of UV inactive small organic molecules by using various chemical stains and appropriate analytical conditions. Synthetic/ natural product extractions using various solvents. Guiding and deciding about an appropriate purification method based on synthetic route for product and the stability of the product. Dealt with sensitive reaction products (in crude), from a few milligrams up to pounds. Spinning band distillation, soxhlet extraction and re-crystallization. Participated in problem solving, by discussion with synthetic organic chemists. Technical maintenance and quick troubleshooting of HPLC and Biotage machines. Used Buchi’s advanced liophilizers/ freeze drying techniques to remove high boiling solvent and acids/ bases (formic acid, TFA, ammonia), from purified products. Cost-cutting working. Submission of finished goods to Quality Control (QC).
