Ce projet de these fait partie d'un projet collaboratif impliquant differentes equipes : celle de F. Roussi (ICSN, Universite Paris Saclay, Gif-sur-Yvette, laboratoire d'accueil), celle de B. Antonny (IMPC, Nice), et la plateforme de criblage pharmacologique CIBI (J. Bignon, ICSN). Le laboratoire d'accueil etudie depuis plusieurs annees une famille de produits naturels, les schweinfurthines (SWs), au potentiel anticancereux prometteur. Les objectifs de ce projet de these sont : 1) d'elaborer une prodrogue de type azobenzene d'un lead derive des SWs permettant la liberation ciblee de sa forme active ; 2) d'elaborer des sondes off-on de type azobenzene afin d'etudier leur cible biologique dans un contexte cellulaire, la proteine OSBP.

The Hepatitis D virus (HDV) is a virus that, by co-infecting hepatocytes with the Hepatitis B virus, leads to severe liver failure and an increased risk of liver cancer. Currently, there is no treatment available for HDV. Recent studies show that ATCase plays a key role in the HDV replication cycle, and that a known ATCase inhibitor, N-(phosphonoacetyl)aspartic acid, could be an effective inhibitor against HDV replication. In this study, we conducted an examination of the structural and mechanistic properties of PALA in ATCase using docking approaches. Based on our observations, a series of substrate analogues was considered and synthesized.
